Sedimentary delta(15)N records from the oligotrophic western equatorial Pacific (WEP) off Mindanao show that late Holocene sedimentary delta(15)N is substantially lower than that of the early Holocene, following a gradual >3 parts per thousand decrease that occurred between 7 and 3 kyrs ago. Analyses of modem day nitrate isotope profiles from the same region indicate the sensitivity of the WEP N pools towards (1) the advection of (15)N-enriched nitrate from the Eastern Equatorial Pacific (EEP) by the North Equatorial Current (NEC) and the Mindanao Current in subsurface waters and, (2) at shallow depths, the input of new and (15)N-depleted nitrate through N, fixation. Modern instruments together with the use of standard reference materials have improved the accuracy and long-term reproducibility of the analysis of nutrients (ammonium, nitrate, nitrite, phosphate and silicate) in sea water using segmented-flow analysis, so that errors arising from matrix effects become more significant. Colorimetric detectors with bubble-through flowcells have become widely used for seawater analysis in recent years and their associated matrix effects are described. In an attempt to synthesize conceptual and experimental information on the behaviour of enteric bacteria in seawater, a mathematical model has been developed. This model is based on changes in intracellular metabolisable components depending an physiological responses of the cell when subjected to nutrient starvation, salinity stress and solar radiation. Following a strategy which takes into account short- and mid-term physiological adaptation and reversal processes, the cells can develop in different states: culturable (B-1), viable and definitively nonculturable (B-2), and reversibly dormant (B-3) cells. Absolute salinity measurement of seawater has become a key issue in thermodynamic models of the oceans. One of the most direct ways is to measure the seawater refractive index which is related to density and can therefore be related to the absolute salinity. Air-sea CO2 flux variability in frontal regions of the Southern Ocean from CARbon Interface OCean Atmosphere drifters ArchiMer

Nine CARbon Interface OCean Atmosphere (CARIOCA) drifters were deployed in the Southern Ocean (south of the subtropical front, STF) between 2001 and 2006. They recorded 65 months of measurements in all seasons between 57 degrees S and 40 degrees S. Hydrological fronts detected by altimetry indicate that one buoy explored the polar zone (PZ) of the Atlantic Ocean and the western Indian Ocean; the remaining buoys explored the northern and southern parts of the subantarctic zone (SAZ) from the mid-Indian Ocean (73 degrees E) to the eastern Pacific Ocean (112 degrees W). An automated photo-oxidation method for the determination of dissolved organic phosphorus in marine and fresh water ArchiMer

A segmented flow automated method with on-line photo-oxidation for the determination of dissolved organic phosphorus + soluble reactive phosphorus (DOP + SRP) in seawater and fresh water is described here. A low-power lamp was used for a compact, easy-to-handle and low-ozone-producing manifold. The influence of seawater matrix components was studied in detail using natural seawater and salt solutions spiked with DOP model compounds. Bromide was found to be the most inhibitory species in seawater. The work shows that most salt solutions referred to as artificial seawater are not satisfactory model matrices to test the actual seawater matrix effect. Analysis of hydrogen and methane in seawater by “Headspace” method: Determination at trace level with an automatic headspace sampler ArchiMer

“Headspace” technique is one of the methods for the onboard measurement of hydrogen (H2) and methane (CH4) in deep seawater. Based on the principle of an automatic headspace commercial sampler, a specific device has been developed to automatically inject gas samples from 300 ml syringes (gas phase in equilibrium with seawater). As valves, micro pump, oven and detector are independent, a gas chromatograph is not necessary allowing a reduction of the weight and dimensions of the analytical system. Determination of iron-porphyrin-like complexes at nanomolar levels in seawater ArchiMer

A new method for the non-specific determination of iron-porphyrin-like complexes in natural waters has been developed. It is based on the chemiluminescent oxidation of the luminol in the presence of dioxygen (O-2) at pH 13. The method has been implemented in a FIA manifold that allowed the direct injection of seawater. The limit of detection is 0.11 nM of equivalent hemin (Fe-protoporphyrin IX). Determination of rare earth elements and other trace elements (Y, Mn, Co, Cr) in seawater using Tm addition and Mg(OH)(2) co-precipitation ArchiMer

This paper reports on a novel procedure for determining trace element abundances (REE and Y, Cr, Mn, Co) in seawater by inductively coupled plasma sector field mass spectrometry (ICP-SFMS). The procedure uses a combination of pre-concentration using co-precipitation onto magnesium hydroxides and addition of thulium spike. The validity of the method was assessed onto 25 ml volumes of certified reference materials (NASS- and CASS-4) and in house seawater standard. Determination of the copper isotope composition of seawater revisited: A case study from the Mediterranean Sea ArchiMer

A new technique for the determination of dissolved copper isotope composition (δ65Cu) of seawater was applied to examine copper sources and internal cycling in the Mediterranean Sea. A succession of chelating resin with nitrilotriacetic acid functional groups and strong base anion exchange resin, together with optimization of the multi-collector inductively coupled plasma mass spectrometry set-up allowed to isolate copper from seawater matrix and to measure the 65Cu/63Cu ratios in seawater with an external precision of 0.06‰ (2 s.d.). Development of a mreB-targeted real-time PCR method for the quantitative detection of Vibrio harveyi in seawater and biofilm from aquaculture systems ArchiMer

Vibrio harveyi is a particularly problematic Gram-negative bacterium because it can form biofilms on aquaculture facility surfaces, leading to resistance of bacteria against antibiotics and water sanitizers. A SYBR Green I quantitative real-time PCR method was developed to detect V. harveyi directly from environmental samples, including seawater and biofilm. Specific primers targeting the mreB gene were designed. Salinity is the essential parameter for ocean dynamics studies. Its definition and using in the equations used to calculate the thermodynamic properties of seawater, were revised in 2010, opening the possibility of new sensors developments. In this context, NOSS (NKE Optical Salinity Sensor) has emerged as one of the first underwater instrument for in situ refractive index measurement in the past years open up the scope of possibilities to access to density parameter. This achievement is the fruit of the cooperation between several institutions and the fruit of several years of developments and trials. NOSS sensor is designed to be deployed in coastal environment and open-ocean waters up to 2000m, especially on profiling floats of the Argo network.
